Effect of shear velocity and flow regimes on scallop post-larval detachment feed on two different diets ArchiMer

Through biological flume experiments, we studied the effect of shear velocity and attachment period on the percent detachment of Pecten maximus post-larvae of different sizes reared on two experimental diets. The first diet, (Pavlova lutheri, Isochrysis galbana, Chaetoceros calcitrans), which served as a reference diet (PTC), is commonly used in bivalve hatcheries; the second is the PTC diet with the addition of Rhodomonas salina (PTCR). Our results indicate that the detachment of post-larvae fed PTCR diet showed significant interactions between scallop length, attachment period and shear stress, while the post-larvae feed with the PTC diet showed only effect of shear stress factor. Effects of age and environment on survival of summer mortality by two selected groups of the Pacific oyster Crassostrea gigas ArchiMer

This study investigated the effects of age (first or second summer of life) and environment (three study sites on French coasts) on summer mortality in Crassostrea gigas, using three generations of oysters produced during the Morest project. For each generation, two groups of oysters, selected either as 'resistant' (R) or 'susceptible' (S) to summer mortality at the age of 6 months in Rivière d'Auray (RA, South Brittany), were evaluated. Three successive generations were produced between 2001 and 2003 to assess to what extent genetic variability exists for survival in juvenile oysters Crassostrea gigas. For each generation, two groups were selected for their high ("R" for resistant) and low ("S" for susceptible) survival. Some positive growth differences can be observed, all year long, in Pecten maximus larvae cultivated in an old fishpond, compared with the water which flows into it periodically. Etude de la nutrition du zooplancton en zone d'upwelling par la mesure des activités enzymatiques digestives ArchiMer

The feeding of the zooplankton has been studied in the natural environment of an upwelling ecosystem by measurements of amylase and protease activities. The hypothesis of enzymatic specific activity as a nutritional index is extended by a correlation with Lorenzen's grazing index. There is no correlation between amylase and protease activity so the diet of the populations is described by the ratio of these two enzymatic activities. Variations of relative feeding rate for Calanus, Temora, Acartia, Podon, and for the total sample are discussed in relation to the ecological and trophic characteristics of the environment.

In the scope of the European program GENEPHYS, which aims to establish the relationship between the genetic base and the physiological characters in relation to the growth in Crassostrea gigas, it has been demonstrated that the most explanatory factors for the growth variability are assimilation performance and aneuploidy. A model linking the assimilation performances and the digestive equipment has been set up and more than 70% of the variability can be explained by two digestive enzymes: laminarinase and amylase. A number of laboratories have decided to focus on the latter.

First results about distribution and changes of biomass (dry weight and total soluble proteins), physiology (amylase, trypsin) and faunistic composition of zooplankton are reported for the two months period following the spill of AMOCO CADIZ on the Britany Northern coast. A shortage of biomass in the aber area is attributed to hydrocarbons. The low level of the mean value of biomasse on the North coasts, and the pecularities of estuarine station of the Lannion bay and of Trieux area are reported and discussed.

Free amino-acids in copepod fecal pellets ArchiMer

By measuring water soluble extracts of free amino acids (DF AA) in the diets and faecal pellets of copepods using high performance liquid chromatography (HPLC), it was found that the molar composition of these molecules in the faeces produced depends on the type of food selected and reflects the hydrolysis of food proteins during digestion. On average, four amino acids (ASP, SER, GLY and LEU) together accounted for 63% of total DF AA in the faecal pellets of a variety of copepod species. The diversity of DFAA in copepod faecal pellets was higher than in sea water or in particles and phytoplankton cells ingested by copepods. Genetic and reproduction strategy in Crassostrea gigas mortality ArchiMer

Bi-parental families were produced in hatchery and tested in the field in 2001. Two sets of 5 families were constituted, selected on their high (R) and low (S) survival. These two sets were reared in Brittany from March to November 2002. Samplings were performed twice a month to obtain data on biometry, survival, reproductive cycle, biochemical composition, adenylate energy charge, hemolymph parameters (ions and defense system) and muscle strength. "R" and "S" oysters exhibited different reproductive effort and spawning strategy. "R" oysters allocated less energy in gonad than "S" ones and presented a complete spawning at the end of August contrary to the "S" which spawns partially. Genetic basis of summer mortality in juvenile cupped oysters ArchiMer

The French project "Morest", coordinated by IFREMER, aims to understand the causes of the summer mortalities in Crassostrea gigas. In 2001, three sets of families were bred following a nested half-sib mating design. 17 half-sib families (HSF) were obtained in this first generation (G1) and reared in 3 sites. Significant differences in survival were observed among HSF, and some HSF showed high levels of mortality in all sites, clearly indicating a genetic basis for survival. Arachidonic acid (20:4n-6, ArA) and its eicosanoid metabolites have been demonstrated to be implicated in immune functions of vertebrates, fish, and insects. Thus, the aim of this study was to assess the impact of ArA supplementation on the FA composition and hemocyte parameters of oysters Crassostrea gigas. Oyster dietary conditioning consisted of direct addition of ArA solutions at a dose of 0, 0.25, or 0.41 mu g ArA per mL of seawater into tanks in the presence or absence of T-Iso algae. Impact of food availability on energy storage and defense related hemocyte parameters of the Pacific oyster Crassostrea gigas during an experimental reproductive cycle ArchiMer

The aim of this study was to test the effect of food quantity on energy storage and defense capacities of oysters during a reproductive cycle. One-year-old Crassostrea gigas oysters were fed two different dietary rations (4% and 12% of oyster dry weight in algal dry weight per day) in controlled experimental conditions over an annual cycle. Oyster dry weights, carbohydrate and lipid contents, energetic adenylate charge, and hemocyte parameters of oysters were significantly affected by reproductive processes related to seasonal temperature variation and, to a lesser extent, by the dietary rations. In vitro modulation of reactive oxygen and nitrogen intermediate (ROI/RNI) production in Crassostrea gigas hemocytes ArchiMer

Bivalve hemocyte competence has been measured by quantifying functional characteristics, including reactive oxygen intermediate (ROI) production after activation with zymosan or phorbol myristate acetate (PMA). However, untreated oyster hemocytes also produce ROI and RNI (reactive nitrogen intermediates) after bleeding even if not stimulated by Zymosan or PMA. Extensive investigation of this parameter by flow cytometry showed that, in vitro, ROI/RNI production by untreated hemocytes maintained in seawater appeared to be independent of both bacterial burden in the serum and non-self particle phagocytosis.
